基于分子分类学对菌株027的鉴定及系统发育分析

摘要 对1株实验室标号027、种名标为红肉拟层孔菌(Fomitopsis rosea)的菌株进行了分子鉴定和系统发育分析,分别以ITS序列与mt SSU rDNA部分序列为目标基因构建了系统发育树,并对相关菌株的ITS-1区与ITS-2区进行了二级结构预测。结果表明,两个系统树均显示菌株027可以与Fomitopsis rosea明显分开而与肉色迷孔菌(Daedalea dickinsii)紧密相连,对ITS-1及ITS-2区的二级结构分析显示菌株027与Daedalea dickinsii的二级结构构型基本相同而能与Fomitopsis rosea明显区分开来,依此为基础,认为菌株027实际上应是Daedaleadickinsii的1株菌株。进一步对Daedalea dickinsii进行系统发育分析认为,Daedalea dickinsii与栓菌属菌物的系统发育关系较远,因此,Daedalea dickinsii不属于栓菌属菌物。 A study was conducted to identify one strain called 027 which is considered as an isolate of a population of Fomitopsis rosea.Phylogenetic analysis of the strain 027 was made by the method of molecular classification.The relationships between the strain 027 and other strains from GenBank were elucidated through comparison of the nuclear internal transcribed spacer(ITS) and the partly mitochondrial small subunit ribosomal DNA(mt SSU rDNA) sequences,and phylogenetic trees were drawn.The secondary structures of the ITS-1 and ITS-2 domains of some species were also predicted.Result shows that the strain 027 could be distinguished clearly from F.rosea and it could clustered into both of the ITS and the mt SSU rDNA trees with Daedalea dickinsii.The secondary structure of ITS-1 and ITS-2 domains also shows that strain 027 is much different from F.rosea.Considering the above results,it is confirmed that the strain 027 is not a strain of F.rosea,it is closely related to Daedalea,and in particular to D.dickinsii.Further research also shows that D.dickinsii is far away from Trametes in phylogenetic trees,so it is considered that D.dickinsii does not belong to Trametes.
