摘要
对白桦花序进行分期取材,CTAB法提取总RNA,基于SMART(Switching Mechanism At5'end of RNATranscript)策略进行cDNA合成,根据现有EST序列设计基因特异性引物,通过RACE(rapid amplification of cDNAend)手段克隆了一花发育相关基因---BplAGL,使用TreeTOP工具对其系统发生进行了评价。采用外标实时定量PCR分析其花期时序表达模式,认为BplAGL可能属于D功能基因。
Total RNA was extracted by CTAB method using the material of Betula platyphylla inflorescence sampled in each developmental stage,and cDNA was synthesized based on switching mechanism at 5' end of RNA transcript strategy.Gene special primers(GSP)were designed according to sequence of an EST in hand.BplAGL,a new gene related to floral development,was cloned by rapid amplification of cDNA ends(RACE),and then its phylogenetic positions were inferred by the tool of TreeTOP.The temporal expression mode of BplAGL during inflorescence development was analysed by quantitative real-time PCR with an outer reference.BplAGL is assumed to belong to D-function gene.
