摘要
目的探讨柴胡疏肝散对夹尾应激大鼠脑和胃GASR mRNA和CCK-AR mRNA表达的影响。方法将30只大鼠随机分为3组,正常对照组、模型组和柴胡疏肝散组,每组10只,采用慢性夹尾刺激方法制作慢性应激模型,分别给予生理盐水、柴胡疏肝散水煎剂灌胃,共4周。观察其一般情况及胃组织学变化,并用原位杂交方法检测大鼠脑和胃组织胃泌素受体(GASR)mRNA和胆囊收缩素受体A(CCK-AR)mRNA的表达。结果①模型组出现了抑郁和消化不良行为表现,而柴胡疏肝散组则较不显著;②模型组胃黏膜均出现不同程度的糜烂和炎症反应,而柴胡疏肝散组则较轻;③模型组大鼠胃和脑组织上的GASR mRNA表达量较正常对照组有显著升高(均P<0.05),柴胡疏肝散组则较模型组有显著降低(均P<0.05)。模型组大鼠胃组织CCK-ARmRNA表达较正常对照组显著升高(P<0.05),柴胡疏肝散组较模型组有显著降低(P<0.05)。结论柴胡疏肝散能改善慢性应激抑郁和消化不良行为,其机制可能与其能减低脑和胃组织GASRmRNA表达和减低胃组织CCK-ARmRNA表达有关。
Objective To study the effects of Chaihushugansan on the gastric receptor mRNA (GASR mRNA) and CCK-A receptor mRNA (CCK-AR mRNA) expression in stress rats induced by constantly squeezing their tails.Methods 30 rats were selected and randomly divided into 3 groups:control group,model group and Chaihushugansan group (each n = 10).The animal model of stress was established by constantly squeezing their tails for 4wk and mean while saline,Chaihushugansan decoction were administered respectively to three groups by gavage.The animal's general situation and pathological change were observed every week,and GASR mRNA and CCK-AR mRNA expression of rats in brain and gastric tissues were measured four weeks later.Results ① The model group appeared depression and dyspepsia,the manifestation in Chaihushugansan decoction may not be so obvious compared with model group.② Model group appeared different moderate stomach mucosa erosion and inflammation but they were not so obvious in Chaihushugansan group.③ The GASR mRNA and CCK-AR mRNA expression of the model group rats in brain and gastric tissues were increased significantly than those of control group(P〈0.05),while Chaihushugansan group was significantly lower than model group(P〈0.05).Model group was significantly higher than control group in CCK-AR mRNA of gastric tissue(P〈0.05).Conclusion Chaihushugansan can improve depression and dyspepsia,its mechanism is related to decreasing expression of GASR mRNA in brain and gastric tissues and CCK-AR mRNA in gastric tissne of rats.
