细胞外信号调节激酶在大鼠胰腺纤维化中作用的研究 被引量:4

STUDY OF EXTRACELLULAR REGULATED PROTEIN KINASES ON PANCREAS FIBROSIS IN RAT

摘要 目的研究细胞外信号调节激酶(ERK1/ERK2)在大鼠胰腺纤维化中的作用。方法通过对大鼠胰管内注射2%三硝基苯磺酸(TNBS)的方法制备大鼠胰腺纤维化模型。采用免疫组化技术检测第四周胰腺组织中α-平滑肌肌动蛋白(α-SMA)、血小板衍化生长因子(PDGF)、磷酸化细胞外信号调节激酶1、2(P-ERK1/P-ERK2)的含量。结果 PDGF、P-ERK1/P-ERK2、α-SMA在模型组的表达均强于对照组(P<0.01)。α-SMA与PDGF、P-ERK1、P-ERK2定量分析成正相关(P<0.05)。结论 ERK1/ERK2信号传导通路在慢性胰腺炎发生纤维化过程中具有重要作用。 Objective To study the expression of ERK1 / ERK2 on pancreas fibrosis in rat. Methods 2% trinitro-benzene-sulfonic acid ( TNBS ) at a concentration was injected into the rats' pancreatic duct to set up the model of pancreatitis; 4 weeks later,the expression of PDGF,α-SMA,P-ERK1 / P-ERK2 were determined in pancreas tissues by using immnnohistochemistry technique. Results Expressions of PDGF,α-SMA,P-ERK1 / P-ERK2 were stronger in the model than that in the control group on the fourth week. There was a positive correlation between α-SMA and PDGF and P-ERK1 / P-ERK2. Conclusion ERK1 / ERK2 are act mainly to play a key role in pancreas fibrosis and its development.
