高效液相色谱法测定滴眼剂中的硝酸毛果芸香碱及其水解产物 被引量:3

摘要 硝酸毛果芸香碱滴眼剂是眼科常用的青光眼治疗剂和缩瞳药。滴眼剂中的毛果芸香碱(Ⅰ)不稳定,在制备、贮存过程中,可水解成毛果芸香酸等水解产物(Ⅱ),导致药物的药理活性降低。 The contents of pilocarpine nitrate and its hydrolysates in eyedrops were determined by RP-HPLC using benzoic acid as internal standard. A nucleosil 10 C18 column (250×2.6mm) was used with methanol-0.1 mol/L acetate buffer (pH4.5) (40:60)as mobile phase. The detection wavelength of UV detector was 230nm. The peaks were identified by adjusting the pH value of the mobile phase. The linear relationship between peak height and the quantity injected (64-320μg/ml) was obtained. The coefficient of Variation was less than 1.0%. It was found that the. quantity of the hydrolysates increased when the preparation was carried out under heating.
