The Determination of Inosine 5'-Monophosphate in Muscle by Hi9h-Performance Liquid Chromatography

肌肉中肌苷酸的含量是肌肉鲜度的一种标志。近年来肌苷酸有watanabe等用多种酶电极法测定,Kitada等用离子对色谱法测定。本文采用反相色谱法对大白鼠和小白鼠腿肌中的肌苷酸用外标法进行定量测定。

A method is presented for the determination of inosine 5-monopuosphate (IMP) in the muscle of mouse and rat by nigh-performance liquid cbromatography on Hypersil C18 column with 40 mmol/L phosphate buffer solution as mobile phase.The quantitative determination was based on external standard method.The coefficient of Variation and recovery were 0.1-1.3% and 97-104% respectively at the detection level of Ca.50 ppm.The minimun detectable amount was 6ng.
