IL-1β体外诱导皮质与中脑腹侧神经干细胞向TH阳性细胞分化差异研究

摘要 目的:探讨IL-1β对鼠胚脑皮质区与中脑区来源的神经干细胞向酪氨酸羟化酶(tyrosine hydroxylase,TH)阳性细胞分化的差异。方法:体外分别分离、培养胚鼠脑皮质区与中脑区来源的神经干细胞并传代,在有血清条件下IL-1β诱导分化,并设空白组对照,分化结果行免疫荧光细胞化学技术鉴定,镜下计数阳性细胞比例。结果:经诱导,中脑神经干细胞在IL-1β作用下分化出TH阳性细胞,其中比例约14%;皮质神经干细胞未分化出TH阳性细胞。结论:中脑神经干细胞在IL-1β作用下可以明显分化出TH阳性细胞,而皮质神经干细胞不能分化,二者体外在向TH阳性细胞分化性能上存在差异。 Objective:To compare cortical neural stem cells(C-NSCs)with mesencephalic neural stem cells(M-NSCs)on the capabilities of differentiation into tyrosine hydroxylase positive (TH-ir) cells in vitro.Methods:The two kinds of NSCs were cultured and passaged in vitro,then IL-1β was respectively used to induce them to differentiate in the condition of DMEM/F12 medium with 10% FBS.Immunocyte chemical techniques and DAPI-cell-count assessment were used to detect the TH-ir cells from two NSCs differentiations.Results: Following IL-1β administration,all M-NSCs groups could be differentiated into TH-ir cells,the proportion was about 14% .However,no TH-ir cells were found in the C-NSCs groups.Conclusion: On the condition of experiment,M-NSCs can significantly be induced to differentiate into TH-ir cells by IL-1β,but C-NSCs can’t.The capabilities of differentiation in vitro of them are different.
