甘蔗叶几丁质酶测定条件的优化

The optimization of determination conditon of chitinase in sugarcane

以新台糖ROC22号甘蔗为材料,采用还原糖比色法测定几丁质酶的酶活,并对酶的反应条件进行优化.结果表明:最佳显色条件为显色剂3,5-二硝基水杨酸用量为1.89mg/ml,显色时间为10min;甘蔗叶几丁质酶的最适反应时间为1h,最适温度为60℃,最适pH值为7.0.

The chitinase activity of sugarcane was determined with DNS method for the determination of reducing sugar.The reaction conditions were optimized.The results indicate that the optimum concentration of DNS reagent was 1.89mg/ml,heating time was 10min;the optimum time of chitinase of sugarcane was 1h,the temperature was 60℃,and the optimum pH was 7.0.
