摘要
[目的]探索影响羊草愈伤组织分化及再生的因素,为羊草组织培养及转基因受体系统建立提供理论依据。[方法]选取5种类型羊草种子进行组织培养,研究在愈伤组织继代、分化培养过程中培养基类型、激素种类和浓度、低温处理等因素对分化率的影响。[结果]不同种群的羊草愈伤组织分化率差异极大;在愈伤组织诱导和继代过程中添加2.0mg/L2,4-D有利于羊草胚性愈伤组织再生能力的保持;继代培养时,不同种类培养基对分化率影响不大,低温处理不能提高长时间继代培养愈伤组织的分化率;分化培养时,外源激素对提高羊草分化率作用不大。[结论]影响羊草愈伤组织分化率的因素主要包括种质资源以及诱导和继代过程中的激素浓度。
[Objective]To explore the factors affecting callus regeneration and differentiation in Chinese wildrye(Leymus chinensis),and provide a theoretical basis for the tissue culture and establishment of transgenic receptor system of Leymus chinensis.[Method]A total of five types of Leymus chinensis seeds were selected,and the effects of media types and hormones,hormones concentration,low temperature on differentiation rate were studied at the process of callus subculture and differentiation.[Result] The differentiation rate of Leymus chinensis was associated with its population.2,4-D(2.0 mg/L)was beneficial to maintain the regeneration capacity of embryogenic callus in the process of callus induction and subculture.There was little effect of media on differentiation.Low-temperature processing could not improve the differentiation rate of subculture callus for long period.Exogenous hormone had little effect on the differentiation rate of Leymus chinensis in differentiation culture.[Conclusion] The factors affecting the differentiation rate of callus mainly include germplasm resources and the hormone concentration in the process of induction and subculture.
