摘要
目的:应用代表迟发型超敏反应的小鼠变应性接触性皮炎模型及体外脾细胞IFN-γ、IL-4的分泌,考察玉屏风散醇提液和乙酸乙酯萃取物抑制迟发型超敏反应的不同特点。方法:用二硝基氯苯(DNCB)诱导小鼠变应性接触性皮炎,考察玉屏风散提取物诱导相和效应相分别给药的干预作用;用ELISA法检测ConA刺激的脾细胞IFN-γ、IL-4的分泌水平。结果:玉屏风散醇提液在诱导相和效应相给药均能减轻小鼠耳肿胀度,改善耳组织病理学改变,体外显著抑制ConA诱导的脾细胞IFN-γ、IL-4的分泌;乙酸乙酯萃取物效应相给药显著减轻小鼠耳肿胀度,改善耳组织病理变化,在诱导相则影响不明显,显著抑制ConA诱导的IFN-γ分泌,一定程度促进IL-4分泌。结论:玉屏风散醇提液和乙酸乙酯萃取物抑制迟发型超敏反应具有不同的特点,且与对T细胞的不同影响有关。
Objective:Yupingfeng powder is a common used formula of Chinese Medicine for regulating immune function. In order to evaluate effects and features of two different extracts (alcohol extract(A) and ethyl acetate extract(B) of the formula) on delayed type hypersensitivity (DTH),their effects on mice allergic contact dermatitis (ACD,a model for DTH),and secretion of the cytokines was examined. Methods:Mice ACD were induced by DNCB. IFN-γ and IL-4 induced by ConA were determined by ELISA. Results:It is showed that the swelling degree and pathological changes of mice's auricle in ACD was reduced significantly by treated with A either in induction phase or effector phase in vivo. IFN-γ,IL-4 secretion stimulated by ConA was inhibited markedly by A in vitro. While the swelling degree of mice's auricle of ACD was reduced by treated with B in effector phase but no significant effect was detected in induction phase. IFN-γ secretion stimulated by ConA was inhibited significantly by B while IL-4 level was increased slightly. Conclusion:The results indicated that A and B from Yupingfengsan inhibit DTH with different function characteristics.
