摘要
目的:探讨栀子总环烯醚萜苷对大鼠脑出血AQP4和MMP-2/9表达的影响。方法:SD大鼠随机分为假手术组、模型组和栀子总环烯醚萜苷(30mg/kg)治疗组,采用自体血注入诱发大鼠脑出血模型,术后24、48、72和120h时间点各组各取5只大鼠脑组织标本,采用干湿重法、免疫组织化学染色、逆转录聚合酶链反应技术分别测定脑含水量、AQP4蛋白和AQP4mRNA的表达、MMP-2和MMP-9表达。结果:与模型组比较,治疗组大鼠在各时间点脑含水量减少(P<0.01),AQP4蛋白和AQP4mRNA表达降低(P<0.05),术后72~120h时间点MMP-2和MMP-9表达水平低(P<0.05~P<0.01)。结论:栀子总环烯醚萜苷可能通过抑制AQP4、MMP-2和MMP-9的表达减轻脑出血后脑水肿的形成。
Objective:To study the influence of total iridoid glycosides in Gardenia jasminoides on the expression of AQP4 and MMP-2/9 in perihematomal brain regions of rats. Methods:Male Sprague-Dawley rats were randomly divided into pseudo-operation group,ICH model group and the treatmengt group of Gardenia jasminoides total iridoid glycosides (30 mg/kg). ICH models were induced in rats by injecting stereotactic infusion autologous blood into pallidus. The brain tissue specments of 5 rats in every group were respectively gathered at the time of 24h,48h,72h and 120h after operation. The expression of AQP4 protein and gene mRNA and MMP-2/9 in rat after ICH were assessed by inmunohistochemistry and reverse trascriptase polymerase chain reation,at the same time the moisture in brain tissue was measured by dry-wet weighting method. Results:Compared with ICH model group,the expression of AQP4 protein and AQP4 mRNA were decreased obviously,and moisture in brain tissue in Gardenia jasminoides iridoids groups(30 mg/kg) were remarkably declined at every time point. The level of MMP-2/9 in treatment group was lower than that in the model group at 72 to 120 hour after ICH. Conclusion:Gardenia jasminoides total iridoid glycosides groups can reduce the expression of AQP4 and MMP-2/9 in rat after ICH,which may be related with the anti-edema effect folloming ICH.
