罗格列酮对2型糖尿病大鼠肾脏的抗氧化保护作用 被引量:3

摘要 目的观察新型胰岛素增敏剂罗格列酮对2型糖尿病(DM)大鼠肾脏的抗氧化保护作用。方法将30只雄性Wistar大鼠随机分为3组:正常对照组、DM组和罗格列酮治疗组,每组10只。药物干预8周后,检测各组大鼠空腹血糖(FBG)、血胰岛素、总胆固醇(TC)、三酰甘油(TG)、血肌酐(SCr)、24h尿白蛋白排泄率(UAE)及肾组织中丙二醛(MDA)含量及铜/锌-超氧化物歧化酶(Cu/Zn-0D)、过氧化氢酶(CAT)、谷胱甘肽过氧化酶(GSH—Px)活性,同时留取肾组织做HE、PAS染色行病理检查。结果与对照组相比,DM组大鼠FBG、TC、TG、UAE均升高(P〈0.05),系膜密度和肾小球基底膜厚度均增加(P〈0.05);与DM组相比,罗格列酮治疗组FBG、TC、TG差异无统计学意义(P〉0.05),SCr、UAE及肾组织MDA含量降低(P〈0.05),肾组织Cu/Zn-SOD、CAT、GSH—Px活性升高(P〈0.05),肾组织病理表现改善。结论罗格列酮对DM大鼠肾脏有保护作用,其机制可能与抑制肾组织氧化应激反应有关。 Objective To observe antioxidative effects of rosiglitazone on kidney in type 2 diabeteie rats. Methods 30 male Wistar rats were divided randomly into three groups:control group, diabetic group and treatment group. After eight weeks, fasting blood glucose(FBG), blood insulin, total cholesterol(TC), triglyeerides (TG), serum creatinine (SCr), urinary albumin excretory rate (UAE) were measured. Malonaldehyde(MDA)and antioxidant enzymes including Cu/Zn superoxide dismutase (Cu/Zn-SOD) ,eatalase(CAT)and glutathione peroxidase(GSH-Px) in renal tissue were measured. In addition, the renal tissue was observed by light microscopy. Results Compared with control group, FBG, TC, TG, UAE were significantly increased in diabetic group (P〈0. 05 ), the density of mesangiurn and the depth of glomerular basement membrane were significantly enlarged (P〈0. 05). The blood glucose and blood lipid had no significant difference between diabetic group and treatment group (P〈 0. 05). Compared with diabetic group,the level of SCr, UAE and renal MDA in treatment group were significantly decreased (P〈0. 05),but the activity of renal Cu/Zn-SOD, CAT and GSH-Px were increased (P〈0. 05). In addition, the renal pathologic changes in treatment group were also improved. Conclusion Rosiglitazone have protective effect on diabetic nephropathy. The mechanism may be correlated with that it inhibite oxidative stress,
