摘要
目的探讨血管平滑肌细胞不同细胞周期中线粒体融合蛋白2(mitofusin2,Mfn2)表达水平的变化。方法通过血清饥饿法、胸腺嘧啶核苷(thymidine)和诺考达唑(nocodazole)干预,使血管平滑肌同步化在不同的细胞周期(G0/G1期、G1/S期、S期、G2/M期);流式细胞仪鉴定细胞周期;Western blot检测Mfn2在不同细胞周期中的表达水平。结果 (1)流式细胞仪鉴定各组DNA含量百分比显示G0/G1期、G1/S期、S期、G2/M期均已同步化,符合本次实验要求。(2)Western blot示:同步化干预后,Mfn2在G0/G1期(静止期)的表达量显著高于G1/S期及S期(增殖期);自然状态下,Mfn2在G0/G1期表达量亦明显高于S期。结论 Mfn2表达量随细胞周期的变化而变化,Mfn2在细胞周期的静止期的表达量明显高于增殖期。
Objective To evaluate the change of mitofusin 2(Mfn2) expression in the different phases of cell cycle of rat thoracic aortic smooth muscle cells (SMC) in vitro. Methods Serum starvation and drug intervention were introduced to synchronize cells in the different phases of cell cycle (G0/G1-phase,G1/S-phase,S-phase,G2/M-phase) and flow cytometry was used for identification. The expression of Mfn2 was detected by Western blot. Results (1) Flow cytometry revealed that cells were synchronized in G0/G1-phase,G1/S-phase,S-phase and G2/M-phase,respectively. (2) Mfn2 expression in G0/G1-phase (stationary phase) was significantly higher than those in G1/S-phase and S-phase (proliferative phase). Conclusion The expression of Mfn2 changes with the processing of cell cycle and is higher in the stationary phase than it in the proliferative phases.
