补肾活血法对肾小球系膜细胞MMP-2及TIMP-2表达的影响 被引量:4

摘要 目的观察补肾活血法组方中药对基质金属蛋白酶-2(MMP-2)、膜型基质金属蛋白酶-1(MT1-MMP)、基质金属蛋白酶组织抑制剂-2(TIMP-2)在高糖培养的肾小球系膜细胞(GMC)中表达的影响。方法将体外培养的GMC分为高糖组(30mmol/L)、正常糖组(5.4mmol/L)和高糖+补肾活血组方中药组。采用酶联免疫吸附法(ELISA)测定GMC上清液中Ⅳ型胶原的分泌;免疫细胞化学法及流式细胞仪(FCM)检测MMP-2、MT1-MMP、TIMP-2的蛋白表达。结果与正常糖组比较,随着培养时间的延长,高糖组细胞培养上清液中Ⅳ型胶原分泌明显增加,MMP-2、MT1-MMP表达减少,TIMP-2表达升高(P<0.01)。补肾活血法组方中药可以上调MMP-2的表达,下调TIMP-2的表达。结论补肾活血法组方中药可以加速细胞外基质降解,减少细胞外基质积聚,从而防治肾小球硬化。
