摘要
目的调查连云港地区汉族无关个体的15个STR基因座(D8S1179、D21S11、CSF1PO、D3S1358、D7S820、TH01、D13S317、D2S1338、D18S51、D16S539、TPOX、vWA、D19S433、D5S818、FGA)多态性,研究其在法医学检验中的应用价值。方法用AmpFlSTR IdentifilerTM五色荧光标记系统对新鲜血样进行15个基因座的复合扩增,用ABI3130XL全自动测序仪对扩增产物进行检测,用GeneMapper软件进行基因分型。结果 15个基因座累积个人识别能力值为0.9999999999,累积非父排除率达99.999%,家系调查符合孟德尔遗传规律。结论含有15个STR基因座的五色荧光标记复合扩增系统适合用于个体识别和亲子鉴定。
Objective The genetic studies of 15 short tandem repeats(STRs) loci in AmpFISTR IdentifilerTM multiplex amplification systems was carried out in Chinese Han population in Lianyungang.Methods DNA samples from 500 unrelated individuals were screened.The 15 loci were D8S1179、D21S11、CSF1PO、D3S1358、D7S820、TH01、D13S317、D2S1338、D18S51、D16S539、TPOX、vWA、D19S433、D5S818 and FGA.The PCR products were analyzed and genotyped by ABI 3130XL Sequencer.Results The combined power of discrimination (DP) was 0.999 999 999 9 and the combined paternity of exclusion(PE) was 99.999%.These 15 loci met the Hardy-Weinberg expectations.Conclusion The AmpFISTR IdentifilerTM multiplex amplification systems was very useful in forensic case investigation.
