摘要
目的:探讨加味温胆汤对D-半乳糖所致衰老大鼠脑自由基及超微结构的影响。方法:SD大鼠随机分为正常组、模型组(ScD-半乳糖48mg/kg/d)、加味温胆汤组(10g/kg.ig,Sc D-半乳糖48mg/kg/d),连续40天,取大脑皮质测定SOD、MDA、GSH-Px、MAO、LPF及透射电镜观察。结果:注射D-半乳糖后,脑组织SOD活性降低(P<0.01),MDA含量及MAO活性增高(P<0.05),而LPF及GSH-Px活性无明显变化(P<0.05)。加味温胆汤则可提高脑组织SOD及GSH-Px活性(P<0.05和P<0.05),降低MDA含量(P<0.05),抑制MAO活性(P<0.01)。超微结构观察表明,注射D-半乳糖后,脂褐素、溶酶体大小形态不规则,线粒体肿胀、变性、嵴断裂、空泡样变,核膜内褶断裂、核仁边移等衰老改变。加味温胆汤则可使上述变性程度减轻。结论:加味温胆汤可延缓D-半乳糖所致的大鼠衰老,抑制脂质过氧化物的形成,提高抗氧化酶的活性,这可能是该方抗脑衰老的作用机理之一。
Purpose: The influence of Jiaweiwendan decoction on brain free radical and ultrastructure of senile rats due to Dgalactose. Method: Dividing SD rats randomly into normal group, model group (Sc D-galactos 48mg/kg/d), decoction Jiaweiwendan group (10g/kg.ig, Sc D-galactose 48mg/kg/d). 40 days later, determining SOD, MDA, GSH-Px, MAO, LPF in cerebral cortex and observing with TEM.
Results: After the injection of D- galactose, SOD activity was decreased (P< 0.01), MDA content and MAO activity were increased (P<0.05), activities of LPF and GSH - PX appeared without change (P> 0.05). Decoction Jiaweiwendan increased the activities of SOD and GSH-PX(P < 0.01, P< 0.05), decreased MDA content (P<0.05), inhibited MAO activity (P< 0.01). The ultrastructure showed that the size and shape of lipofuscin and lysosome were imegulat, mitochondria were swellon, degenerated, crest broken and vacuolated; nuclear membranes were infolded and broken; mecleoli were margjnated. Decoction Jiaweiwendan could slow down the above changes.
All these suggested that Jiaweiwendan Decoction could delay the senility of rat due to D - galactose, inhibit the formation of lipid peroxidation, increase the activities of antioxidase, and it may be one of the mechanisms of antisenility of the formula.
