红花酢浆草花粉萌发及贮藏特性的研究 被引量:2

摘要 以红花酢浆草(Oxalis corymbosa DC.)花粉为试材,利用离体培养法、碘-碘化钾染色法、醋酸洋红染色法和TTC(氯化三苯基四氮唑)染色法对花粉的生活力进行了测定,在扫描电子显微镜下探讨了花粉形态结构与花粉萌发之间的关系,并研究了不同贮藏条件对花粉萌发的影响。结果表明,花粉培养基中添加不同浓度的蔗糖和硼酸对花粉的萌发有促进作用,花粉萌发的最适培养基为15%蔗糖+0.004%硼酸,其萌发率达31.45%。花粉萌发率与饱满花粉占花粉总数的百分率一致。花粉生活力随贮藏时间的延长而下降,不同的贮藏条件对其有显著的影响,最佳贮藏温度为-80℃,此条件下花粉的耐贮藏力长达30d,贮藏28d后花粉萌发率仍可达到1.30%。 Take the fresh pollen of Oxalis corymbosa as experiment material, germination in vitro, I2-KI staining, carmine acetate dyeing and TTC staining methods were used to determine the ability of the fresh pollen. Effects of different storage conditions on the pollen germination were studied. The results showed that culture media with different concentration of sucrose and boric had good effects on the pollen germination. The pollen germinating percentage reached the highest after incubated on the media supplemented with 15% sucrose and 0.004% boric acid. The pollen germinating percentage was consistent with the percentage of plump pollens. Vitality of pollen was gradually descended as the storage time lasted; it was the best methods to maintain vitality of pollen at -80℃. The pollen preserved at -80℃ , the storage lasted for 30 d. After 28 d, the rate of pollen germination was 1.30%.
