摘要
从某鸡场病鸡的组织病料中分离出4株疑似多杀性巴氏杆菌的菌株,根据已报道的巴氏杆菌特异序列设计引物进行PCR检测,同时对这些菌株进行细菌生化鉴定,动物接种试验和药敏试验。结果表明,这4个菌株均扩增出一条1200bp左右的DNA带,与预期的大小一致。生化试验结果也与巴氏杆菌的理化特征吻合。说明本试验所设计的引物在鉴定巴氏杆菌病时具有很高的特异性和灵敏度,可作为该病诊断的可靠方法加以推广和应用。动物致病性试验和药敏试验的结果表明,该巴氏杆菌菌株有较强的致病性,且都对先锋噻肟高度敏感。
Four suspected strains of Pasteurella multocida (PM) were isolated from the tissue of sick chickens in a farm. They were detected by PCR with the specific primers, which were designed according to the sequence of PM published before, and were identified by the biochemical test of bacteria. Meanwhile, antibiotic sensitivity test and animal infection test were performed with those strains. Results showed: An DNA band of about 1 200 bp was amplified by PCR with the four strains of PM. This size agreed that of expected amplified product with the special primers. The result of biochemical test was also confirmed that they were Pasteurella multocida. The results showed that the primers designed in this test had very high specificity and sensitivity in the detection of Pasteurella multocida. The method was reliable and was recommended to be used in the diagnosis of Pasteurella disease. It would shorten the time and 1 improve the efficiency of diagnosis. Antibiotic sensitivity test and animal infection test showed that those strains of Pasteurella multocida had stronger pathogenicity, and were highly sensitive to Cefotaxime Sodium.
