摘要
为研究油田产出液中烃降解菌的多样性,设计了特征性烃降解基因的引物,并经基因库回检验证,采用PCR-DGGE方法并结合化学方法分析了油田产出液中烃降解基因及其化学成分。结果表明,产出液中存在丰富的烃降解菌,8个样本共有12种典型的alkB基因类型;其中2种主要的烃降解菌与Bacillussp.BTRH40、Pseudomonas aeruginosaUMI-88这一族群亲缘关系接近;烃降解菌的多样性与样本所处区块的环境条件密切相关。设计的引物及PCR-DGGE方法可方便检测油田产出液的烃降解菌,为油田开发中激活烃降解菌提供了检测方法。
In order to study the diversity of hydrocarbon-degrading bacteria(HDB)in produced fluid of oilfields(PFO)a primer for hydrocarbon-degrading gene(HDG)was designed and proved through GenBank data,PCR-DGGE method was adopted combined with chemical methods to analyze HDG and its chemical components in PFO.The results showed,there was abundance of HDB exists in PFO,and there were 12 kinds of typical alkB gene types totally in 8 samples;among them two main kinds of primary HDB were closely osculated with Bacillus sp.BTRH40 and Pseudomonas aeruginosa UMI-88 these groups of bacteria;And the diversity of HDB was closely related to the location of lands where the samples were collected.Suggested the designed primer and PCR-DGGE method could conveniently detect HDB in the PFO and provide a detecting method to activate HDB in oil field development.
