木立芦荟组织培养及植株再生的研究 被引量:3

摘要 以木立芦荟的茎尖为外植体,接种于不同激素配比的MS固体培养基上,通过直接器官发生和间接器官发生两种方式获得再生植株。试验结果表明:直接诱导不定芽的最佳诱导培养基为MS+6-BA4.0mg/L+NAA0.1mg/L,平均芽数可达3.67;间接诱导过程中,愈伤组织诱导的最佳培养基是MS+BA2.0mg/L+IBA0.2mg/L,诱导率为100%;不定芽生根的最佳培养基为1/2MS+NAA0.1—0.2mg/L;在培养基中添加0.2%的活性炭可有效抑制外植体褐化;组培苗炼苗20余天,将其盆栽,成活率可达80%。 With the shoot of Aloe arborescens Mill.as explants,MS mediums with different concentration of plant hormones were used to tissue culture and plant regeneration through direct and indirect organogenesis.The results showed that the optimum formula of medium for adventitious buds inducement was MS+6-BA 4.0mg/L+NAA 0.1mg/L,and the average number of buds was 3.67.During indirect organogenesis,the most ideal medium of callus inducement was MS+BA 2.0mg/L+IBA 0.2mg/L,the induction rate was 100%.The best medium of rooting was 1/2MS + NAA 0.1—0.2mg/L.Adding 0.2% active carbon could inhibit the browning of explants.The plantlets were exercised for 20d and moved to pots,and the survival rate was 80%.
