不同剂量左旋精氨酸对骨质疏松大鼠骨代谢的作用 被引量:1

The Effect of Bone Metabolism on Different Dosage L-arginine to the Osteoporosis Rats

摘要 目的探讨不同剂量左旋精氨酸(L-Arg)对骨质疏松大鼠血清一氧化氮和骨显微结构变化的作用。方法将50只3月龄雌性大鼠随机分为假手术组(Sham组)10只和模型组(OVX)40只,对模型组进行去势制造骨质疏松模型,12周后随机抽取10只进行骨密度测量,证实骨质疏松模型建立成功后,再随机4组:OP组,LL-Arg组、ML-Arg组和HL-Arg组,每组10只,OP组正常喂养,其他三组分别以不同剂量的L-Arg皮下注射。12周后测量各组大鼠腰椎骨密度,下腔静脉取血测量血清NO水平,右侧股骨组织形态学观察。结果三组不同剂量L-Arg治疗12周后:①OP组大鼠血清NO水平比Sham组明显降低(P<0.05);LL-Arg组、ML-Arg组和HL-Arg组大鼠均明显高于OP组(P<0.05,P<0.01,P<0.01);②OP组BMD相比Sham组明显降低(P<0.05);LL-Arg组、ML-Arg组大鼠明显高于OP组(P<0.05);HL-Arg组大鼠与OP组相比差异无显著性(P>0.05);③光学显微镜观察LL-Arg、ML-Arg组和Sham组在骨小梁数量、连续性等方面均明显优于OP组,而HL-Arg组则与OP组相当。结论切除双侧卵巢可诱导绝经后骨质疏松模型;外源性精氨酸可提高血清NO水平;适当浓度NO可提高绝经后骨质疏松大鼠骨密度,增加骨小梁数量、连续性,防治绝经后骨质疏松;高浓度NO虽可提高血清NO水平,但不能提高骨密度,不能阻止去卵巢大鼠骨质退化,骨量丢失。提示临床上应用低、中剂量L-Arg防治绝经后骨质疏松症。 Objective Explore different dose L-Arginine(L-Arg) to osteoporosis rats serum nitrogen monoxidum(NO) and bone micro-structural changes in the role.Methods 50 cases of 3-month-old female rats will randomly divide into two group: 10 sham-operated group(sham group) and 40 ovariectomy group(ovx group).Making model to osteoporosis model group using the method of resection ovaries,after 12 weeks randomly selected 10 to measure of bone mineral density(BMD),to confirm the osteoporosis model is successful,then randomly divide into 4 group: osteoporosis-group(OP group),low-dose L-Arginine-group(LL-Arg group),medium-dose L-Arginine-group(ML-Arg group),high-dose L-Arginine-group(HL-Arg group),hypodermic injection with different doses of L-Arg;After 12 weeks,Measure of lumbar spine bone mineral density,to measure the value of serum nitrogen monoxidum from the inferior vena cava blood,Histological observation of the right femur.Results Three different doses of L-Arg to treat the rats after 12 weeks: ①OP group of serum NO levels significantly lower than in Sham group(P〈0.05);LL-Arg group,ML-Arg group and HL-Arg group rats were significantly higher than the OP group(P〈0.05,P〈0.01,P〈0.01);② OP group BMD compared with Sham group were significantly lower(P〈0.05);LL-Arg group,ML-Arg group rats was significantly higher than OP group(P〈0.05);HL-Arg group rats compared with the OP group was no significant difference(P〉0.05);③Optical microscopy LL-Arg group,ML-Arg group and the Sham group trabecular bone volume,continuity,etc.were significantly better than OP group,while the HL-Arg group and the OVX group were quite.Conclusions Ovariectomia bilateral can induce model of postmenopausal osteoporosis;exogenous L-arginine can increase serum NO levels;the right consistency NO can increase bone mineral density in postmenopausal osteoporosis and increase trabecular bone volume,continuity,prevention and treatment postmenopausal osteoporosis;Although high consistency NO can increase serum NO levels,but can not increase bone density,can not stop the degradation of bone in ovariectomized rats,bone mass loss.Prompted use the clinical application of low-,medium-dose L-Arg to prevent and treat postmenopausal osteoporosis.
