摘要
采用碱-酶结合法以人胎盘为原料提取胶原蛋白,并使用原子力显微镜(AFM)、差示扫描量热仪(DSC)、圆二色光谱(CD)和傅立叶变换红外光谱(FTIR)对所得胶原蛋白产品进行表征。结果表明:此工艺路线的胶原蛋白得率高,达到0.102%;提取时间由48 h以上减少到24 h以内,而且所得胶原蛋白分子完整地保留了其螺旋纤维结构。此法是从人胎盘中提取胶原蛋白的一种行之有效的方法。
The collagen was extracted from the human-placenta by a alkali-enzyme method.The properties of the collagen were investigated by atomic force microscope(AFM) and differential scanning calorimeter(DSC),circular dichroism(CD),and Fourier transform infrared spectroscopy(FTIR).The results indicated that the collagen yield was 0.102% by the new method.The extraction time was reduced from 48 h to 24 h.In addition,the collagen molecules held the fibrillar structure.The new method was effective for extracting the collagen from human-placenta.
