摘要
目的:探讨不同脑温状态下银杏叶提取物(GBE)对大鼠脑缺血再灌注损伤的影响。方法:将大鼠随机分为常温假手术组、常温脑缺血对照组、亚低温脑缺血组、轻度高温银杏叶组、常温银杏叶组、亚低温银杏叶组,测定各组大鼠缺血脑组织SOD、GSH-Px、MDA含量,采用光镜观察各组大鼠缺血脑组织病理改变特点。结果:亚低温银杏叶组、亚低温脑缺血组、常温银杏叶组、常温脑缺血对照组、轻度高温银杏叶组大鼠缺血脑组织病理损伤呈依次加重趋势,与SOD、GSH-Px、MDA水平失衡有关。结论:①GBE对脑缺血的保护作用受不同脑温影响,37~37.5℃常温状态下GBE的神经保护作用有限,与脑温较高时GBE对氧自由基的清除能力有一定限度有关,而32~32.5℃亚低温状态下GBE对脑缺血的保护作用增强,与亚低温状态下GBE对氧自由基的清除能力增强有关。②亚低温和GBE联合干预,增强清除氧自由基的能力及增强对脑缺血的保护,可能不是单一因素的作用,而是亚低温作用为主,两种干预因素共同协同作用的结果。
Objective:To study the effect of Ginkgo biloba extract(GBE)under different cerebral temperature on the cerebral ischemia/reperfusion injury of rats.Method :The rats were randomly divided into 6 groups,including the sham operated group in normal temperature,cerebral ischemia group in normal temperature,cerebral ischemia group in subhypothermia,GBE group in mild hypothermia temperature,GBE group in normal temperature,GBE group under subhypothermia temperature.Then the concentration of SOD,GSH-Px,and MDA were detected in variant ischemia brain group.The light microscope and electron microscopy were used to observe the pathology change of cerebral tissue in rat focal cerebral ischemia under different cerebral temperature.Results :The pathological damage rang and extent of cerebral tissue were becoming more seriousier in GBE group under subhypothermia temperature,cerebral ischemia group in subhypothermia,GBE group in normal temperature,cerebral ischemia group in normal temperature,GBE group in mild hypothermia temperature respectively.The pathological damage was relevant to the disbalance of the concentration of SOD,GSH-Px,and MDA.Conclusion :①During the processes of cerebral ischemic,different brain temperature may affect the effect of GBE's neuroprotection.This limited neuroprotective effect was relevant to the decreased cleaning oxygen free radical of GBE under normal temperature(37 ~ 37.5℃),and was enhanced neuroprotective effect was relevant to the increased cleaning oxygen free radical of GBE under subhypothermia(32~32.5℃).②It suggests that the enhanced effect of cleaning oxygen free radical and neuroprotection in focal cerebral ischemia tissue of GBE group under subhypothermia isn't the result of either subhypothermia or GBE intervention factor,but the main effect of subhypothermia and cooperation of both.
