高效液相色谱法测定痫乐散中苯巴比妥含量

RP-HPLC Determination of Phenobarbital Content in Xianlesan

目的:建立高效液相色谱法测定痫乐散中苯巴比妥的含量。方法:采用Diamonsil(钻石)C18色谱柱(5μm,250mm×4.6mm);以甲醇-磷酸盐缓冲液(磷酸二氢钾6.8g,加三乙胺4mL,加水至1000mL)(47:53)为流动相;流速:1.0mL/min;检测波长:215nm;进样量:20μL。结果:苯巴比妥线性范围为6～60μg/mL,r=0.9999;平均回收率为99.9%,RSD=0.52%,n=9。结论:该方法简便准确,专属性好,可以作为痫乐散中苯巴比妥含量的测定方法。

Objective：To establish an RP-HPLC method for the determination of phenobarbital content.Method ：Uses Diamonsil the C18 Chromatographic column（5μm,250mm×4.6mm）.The mobile phase consisted methanol-phosphate solution（potassiun dihydrogen phosphate 6.8g,triethylamine 4mL,water to 1000mL）（47 ：53）,the flow rate was 1.0mL/min.The detection wavelength was 215nm.Inject volumn was 20μL.Results ：The linear range for the content of phenobarbital was 6～60μg/mL ;the coefficient was 0.9999.The average recovery of phenobarbital was 99.70% and its RSD was 0.57%（n=9）.Conclusion ：This method simple accurate,the specificity is good,may be used for assay of the phenobarbital content in xianlesan.
