毛葡萄叶水提物对大鼠脑缺血再灌注的保护作用 被引量:2

摘要 目的探讨毛葡萄叶水提物对SD大鼠脑栓塞后缺血再灌注的脑保护作用及其可能作用机制。方法连续灌胃给药8d后建立SD大鼠脑栓塞模型,作TTC脑染色,观察脑梗死面积;HE染色观察脑梗死区的形态学变化;用免疫细胞化学检测脑组织相关凋亡基因bcl-2及caspase-3的表达变化;并检测大鼠血浆丙二醛(MDA)、一氧化氮(NO)及乳酸脱氢酶(LDH)水平变化。结果毛葡萄叶水提物能明显减小大鼠脑栓塞模型的梗死面积,明显提高bcl-2的表达和下调caspase-3的表达,并有降低血浆MDA水平和升高LDH、NO水平的作用,且毛葡萄叶水提物组的效果优于阳性药物对照组。结论毛葡萄叶水提物有明显抗脑梗死形成作用,其机制可能与抑制细胞凋亡和抗氧化作用有关。 Objective To investigate the protective effect and mechanism of Vitis quinquangularis Rehd water extract(VRE)on SD rat brain after cerebral ischemia-reperfusion.Methods After gastric gavage of VRE for 8 days,SD rat models of cerebral embolism were established.Cerebral pathological changes were observed by TTC staining,and mor-phological changes in cerebral infarction were observed by HE staining.The protein expression of apoptosis-related genes including bcl-2 and caspase-3 was detected by immunocytochemical method.The changes of plasma malondi-aldehyde(MDA),nitric oxide(NO)and lactate dehydrogenase(LDH)levels were also examined.Results Vitis quin-quangularis Rehd water extract significantly inhibited cerebral infarction area in rats with cerebral embolism,increased the expression of bcl-2,down-regulated caspase-3 expression,decreased plasma MDA level and increased LDH and NO levels,the effect being superior to that of the positive control medicine.Conclusion It is suggested that VRE has an obvious effect on prevention of cerebral infarction,and its mechanism may be associated with apoptosis inhibition and anti-oxidization.
