狭基线纹香茶菜对小鼠免疫性肝损伤的保护作用 被引量:6

摘要 目的研究狭基线纹香茶菜对卡介苗(BCG)加脂多糖(LPS)所致小鼠免疫性肝损伤的保护作用。方法 72只小鼠随机分为正常组,模型组,高、中、低剂量给药组(20,10,5g·kg-1),阳性对照组(联苯双酯60mg·kg-)1,每天给药1次,连续12d。第1天,除正常组外,各组每只小鼠尾静脉注射BCG0.2mL(10mg·mL-)1。末次给药后,除正常组外,各组小鼠尾静脉注射LPS0.2mL(37.5μg·mL-1)造模。造模16h后取血,测定血清谷丙转氨酶(ALT)、谷草转氨酶(AST),肝组织超氧化物岐化酶(SOD)的活性和丙二醛(MDA)及一氧化氮(NO)含量,同时测体重、肝脏重、脾重和胸腺重,求脏器指数,并观察肝组织病理学变化。结果狭基线纹香茶菜高、中、低剂量组均能显著降低ALT、AST值和MDA含量(P<0.05或P<0.01),对肝重量和脾重量增加及胸腺萎缩有抑制作用(P<0.05或P<0.01)。病理检查结果也显示有明显的保肝作用。结论狭基线纹香茶菜对BCG和LPS所致小鼠免疫性肝损伤有保护作用,其保肝机制与其抗脂质过氧化损伤有关。 Objective To study the protective effects of Isodon lophanthoides var. gerardianus (Benth.) H. Hara (ILVG) aqueous extract on immunologic liver injury induced by Bacillus Calmette-Guerin vaccine(BCG)and lipopolysaccharide(LPS)in mice.Methods Seventy-two mice were randomly allocated to normal group,model group,low-,middle-and high-dosage ILVG(in the dose of 20,10 and 5 g·kg^-1 respectively)groups,and bifendate(60 mg·kg^-1) group.On the 1st day,except for the normal group,BCG(2 mg for each mouse)was injected into the mice tail intravenously.The mice in the medication groups received the corresponding medicine according to the experimental design by gastric gavage daily for 12 consecutive days.On the 12 th day after the last medication,LPS(7.5μg·mouse-)1was injected into the mice tail intravenously.Sixteen hours later,the serum levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST)were examined.Super oxide dismutase(SOD)activity,malondialdehyde(MDA)and nitric oxide(NO)content in liver tissue were determined.The body weight,and the weight of liver,spleen and thymus of each mouse were measured.Meanwhile,the pathological changes of liver tissue was observed.Results ILVG decreased serum levels of ALT and AST,and reduced MDA content in liver tissue(P〈0.05 or P〈0.01),decreased the weight of liver and spleen and inhibited the shrinkage of thymus(P〈0.05 or P〈0.01),and relieved the hepatic necrosis in pathological observation.Conclusion ILVG aqueous extract possesses the protective effect on immunologic liver injury induced by BCG and LPS in mice,and its mechanism may be through the way of antilipoperoxidation.
