山楂叶总黄酮对人类单核肿瘤细胞增殖的影响 被引量:4

摘要 目的研究山楂叶总黄酮(HF)对人类单核肿瘤细胞U937增殖的影响。方法 U937细胞以5×104个/mL的浓度,培养于10%胎牛血清(FCS)的RPMI1640全培养液中,分别加入浓度为0,25,50,100,150,200mg/L的HF。每组8孔接种于24孔培养板中孵育7d。每天计算各孔细胞密度,第7天测定细胞悬液中乳酸脱氢酶(LDH)活性。结果 HF25~200mg/L组U937细胞密度均低于HF0mg/L组,浓度越高细胞密度越低(P<0.05)。HF0~150mg/L组细胞悬液LDH活性无统计学意义(P>0.05),HF200mg/L组LDH与前5组相比均有统计学意义(P<0.05)。结论 HF具有抑制U937细胞增殖的作用,高浓度的HF(≥200mg/L)对U937细胞可能具有毒性作用。 Objective To study the effect of hawkthorn leaf flavone(HF)on the proliferation of human leukemia U937 cell line.Methods U937 cells(5×10^4/m)lwere cultured in RPMI1640 with 10%fetus cattle serum(FCS)added,and were divided into 6 groups according to the concentration of HF(0,25,50,100,150,and 200 mg/L respectively).Every group was incubated in the 24-well plate for 7 days.Cell count was carried out every day and the activity of lactate dehydrogenase(LDH)in the culture medium was measured on the 7 th day.Results The density of U937 cells in HF 25-200 mg/L groups was significantly lower than that in HF 0 mg/L group(P〈0.05).The activity of LDH in HF 0-150 mg/L groups had no significant difference(P〉0.05),but was increased in HF 200mg/L group as compared with other HF groups(P〈0.05).Conclusion HF has inhibitory effect on the proliferation of U937 cells,and high concentration of HF(≥200 mg/L)probably has cytotoxic effect on U937 cells.
