李斯特杆菌热休克蛋白菌苗免疫原性的研究

摘要 目的:通过免疫动物,以证实李斯特杆菌热休克菌苗有更强的免疫原性。方法:对Ⅰ、Ⅱ、Ⅲ,三组昆明小鼠分别免疫注射李斯特杆菌45℃热休克菌苗、李斯特杆菌37℃常规菌苗和生理盐水,并于第1、7、21天各免疫三次,第28天对小鼠眼底静脉取血,测定中性粒细胞的吞噬率和吞噬指数,试管凝集法测抗体效价,并进行杀菌实验及活菌攻击实验。结果:其中吞噬率、吞噬指数及对李斯特杆菌的抗体效价三项指标,三组均有显著性差异(P〈0.05),以Ⅰ组最高,Ⅱ组次之,其免疫血清对金葡菌亦有抗体效价。杀菌实验两实验组与对照组有显著性差异(P〈0.05),但二实验组间无显著性差异(P〉0.05)。活菌攻击实验,三组其最终死亡率均为100%,无差异性。结论:注射45℃热休克菌苗组其免疫原性较37℃组有所增强。尚有一定的差异性。 Object Through immunization animals to verify that the heat shock vaccine of Listeria Monocytognes(LM) has stronger immunogenicity. Methods The kuming mice were divided into three groups to be immunized three times seperately with 45℃ heat shock vaccine, 37℃ routine vaccine of LM and physiological saline water,respectively, on the lth,Tth,21th day. Drawing the eyeground vein blood on the 28th day to test the neutrophil phagocytic rate ,phagocytic in- dex and the antibody filer by the means of lest tube agglutinin. And carried on the sterilization experiment and living bacteria attack experiment. Result Among the three groups,phagocytic rate,phagocytic index and LM antibody tiler were significantly different( P 〈0.05). Ⅰ group was the highest and Ⅱ group was second . There were also antibody tiler to the Staphloclccus aureus in the immune serum. About the Sterilization experiment,there were significant difference between the two experimental group and control( P 〈 0.05 ), but there was no significant difference between the two experimental group( P 〉 0.05). Living bacteria attack experiment : The last death rates were all 100% ,no difference. Conclusion : The group which was injected 45℃ heat shock vaccine of LM has stronger immuno- genicity than 37℃ group to some extent,and there is a certain degree of difference between them.
