GPI-PLD的表达在甲状腺癌诊断中的临床意义

摘要 目的:探讨甲状腺癌患者糖基化磷脂酰肌醇特异性磷脂酶D(GPI-PLD)的活性变化,为甲状腺癌患者的诊断提供依据.方法:利用具有糖基化磷脂酰肌醇(GPI) 结构的胎盘碱性磷酸酶(PLAP)做底物,通过TX-114分相,定量检测外周血单个核细胞和血浆中GPI-PLD活性,并采用荧光定量PCR外周血单个核细胞中GPI-PLD mRNA水平,分析其表达量与甲状腺癌之间的关系.结果:甲状腺癌患者外周血单个核细胞和血浆中GPI-PLD酶活性分别为(24.15±2.33)%和(29.71±1.52)%,明显低于正常外周血中GPI-PLD酶活性(P<0.001),且甲状腺癌患者单个核细胞GPI-PLD活性仅为其血浆中酶活性水平的81.29%,二者之间有显著性差异(P〈0.05);甲状腺癌患者外周血单个核细胞中GPI-PLD mRNA表达平均值为(0.46±0.17)×10^3copy/ng total RNA,明显低于正常人外周血单个核细胞中的表达水平(1.35±0.41)×10^3copy/ng total RNA (P<0.001).结论:GPI-PLD在甲状腺癌患者中活性及表达明显降低,有可能作为甲状腺癌诊断的指标之一. Objective Discussion of thyroid carcinoma in patients with advanced glycation phosphatidylinositol- specific phospholipase D (GPI- PLD) activity changes to provide a basis for judging the prognosis. Methods With the use of advanced glycation phosphatidylinositol (GPI) structure of the placental alkaline phosphatase (PLAP) substrate to do through the TX-114 phase, the quantitative detection of peripheral blood mononuclear cells and plasma GPI-PLD activity and FQ-PCR using peripheral blood mononuclear cells in GPI-PLD mRNA level of analysis of their expression and the relationship between thyroid carcinoma. Results Patients with thyroid carcinoma in peripheral blood mononuclear cells and plasma GPI-PLD activity were (24.15± 2.33) % and (29.71 ± 1.52) %, significantly lower than in the peripheral blood GPI-PLD activity ( P 〈0. 001), and patients with thyroid carcinoma mononuclear Cell GPI-PLD activity was only the plasma level of activity of 81.29 percent, between the difference was significant ( P 〈0.05) ; Patients with thyroid carcinoma in peripheral blood mononuclear cells GPI-PLD mRNA expression for the average of(0.46 ±0. 17) × 10^3eopy/ng total RNA, significantly lower than the normal human peripheral blood mononuclear ceils of expression(1. 35±0.41) × 10^3copy/ ng total RNA ( P〈0. 001). Conclusions GPI-PLD activity in thyroid carcinoma patients and significantly reduced the expression, it is possible to determine the prognosis of thyroid carcinoma as one of the indicators.
