薄层色谱法快速筛查降脂、降压、止咳平喘类中药制剂中的29种添加化学药物 被引量:39

Detection of 29 Adulterations in Lipid-Regulating,Antihypertensive,Antitussive and Antiasthmatic Herbal Remedies by TLC

摘要 目的建立快速筛查降脂、降压、止咳平喘类中药制剂中非法添加化学药物的薄层色谱法。方法样品用甲醇超声提取,采用硅胶GF254薄层板,降脂类药物以正己烷-乙酸乙酯-乙醚-冰醋酸(10∶8∶2∶1)为展开剂,紫外254 nm检视,并以5%香草醛硫酸试液为显色剂测定;降压类药物以乙酸乙酯-甲醇-二乙胺(40∶0.5∶3)为展开剂,于紫外254、365 nm下检视;止咳平喘类药物以异丙醇-乙酸乙酯-氨水(15∶25∶1.5)为展开剂,于紫外254 nm下检视。结果上述方法可快速筛查出中成药中可能非法添加的10种降脂药物烟酸、普伐他汀钠、瑞舒伐他汀钙、阿托伐他汀钙、氟伐他汀钠、苯扎贝特、辛伐他汀、洛伐他汀、非诺贝特、普罗布考;9种降压药物苯磺酸氨氯地平、盐酸特拉唑嗪、盐酸哌唑嗪、甲磺酸多沙唑嗪、硝苯地平、非洛地平、尼群地平、尼莫地平、拉西地平;10种止咳平喘药物茶碱、二羟丙茶碱、特布他林、羟乙基茶碱、盐酸二氧丙嗪、富马酸酮替芬、咖啡因、盐酸克仑特罗、多索茶碱、盐酸氨溴索等共29种化学药物。结论本方法简便快速,可作为非法添加日常监督检验的有效方法 。 OBJECTIVE To establish TLC methods for fast detection of lipid-regulating agents,antihypertensive agents and antitussive and antiasthmatic agents in herbal remedies simultaneously.METHODS The sample was extracted with methanol and separated by TLC plates pre-coated with silica gel GF254 as stationary phase.Lipid regulating agents were eluted with the mobile phase consisting of n-hexane-ethyl acetate-diethyl ether-glacial acetic acid(10∶8∶2∶1),determined at UV 254 nm and stained with 5% vanillin sulfuric acid solution.Antihypertensive agents were eluted with the mobile phase consisting of ethyl acetate-methanol-diethylamine(40∶0.5∶3) and examined UV 254 nm and UV 365 nm.Antiasthmatic agents were separated with the mobile phase consisting of isopropyl alcohol-ethyl acetate-ammonia(15∶25∶1.5) and examined at UV 254 nm.RESULTS Nicotinic acid,pravastatin sodium,rosuvastatin calcium,atorvastatin calcium,fluvastatin sodium,bezafibrate,simvastatin,lovastatin,fenofibrate,probucol;amlodipine besylate,terazosin hydrochloride,parazosin hydrochloride,doxazosine mesylate,nifedipine,felodipine,nitrendipine,nimodipine,lacidipine;Theophylline,diprophylline,terbutaline,oxyphylline,dioxopromethazine,ketotifen fumarate,caffeine,clenbuterol hydrochloride,doxofylline,ambroxol hydrochloride were detected respectively.CONCLUSION The methods are fast,sensitive and easy to operate,which are fit to be applied for routine test of illegal adulteration.
