白介素5和趋化因子eotaxin在鼻息肉中的表达及其意义 被引量:5

摘要 目的:研究白介素5和趋化因子eotaxin在鼻息肉中的表达,及与嗜酸性粒细胞(EOS)浸润状态的相关性。方法:运用逆转录-多聚酶链反应(RT-PCR)结合半定量分析方法检测20例鼻息肉和8例下鼻甲黏膜组织(对照组)中白介素5和eotaxin的mRNA的表达水平。同时运用HE染色、免疫组织化学方法检测两组中EOS的分布及白介素5和eotaxin的蛋白表达情况,比较两组间差异,分析每组中EOS浸润数目与白介素5和eotaxin表达的相关性。结果:120例鼻息肉组织中以EOS浸润阳性(+)以上者有75%(15例),明显高于8例对照组的12.5%(1例)。且鼻息肉组织中的浸润EOS率21.64±4.33明显高于对照组1.15±0.68(P<0.01);2鼻息肉组织中白介素5和eotaxin的蛋白表达阳性细胞数明显高于对照组(P<0.01);3白介素5和eotaxin的mRNA在鼻息肉组织中的表达明显高于正常对照组(P<0.05)。4白介素5和eotaxin的表达与EOS浸润相关(P<0.01),且两种因子之间也呈正相关关系(P<0.05)。结论:从基因水平和蛋白水平均表明在鼻息肉组织中白介素5和eotaxin含量明显升高,而且与组织中EOS浸润呈明显相关性,提示白介素5和eotaxin是鼻息肉病变中重要的EOS趋化因子,并增强该细胞的局部作用,参与了鼻息肉的发病机制,且二者有协同作用。 Objective: To study the correlation between the expression of IL-5, eotaxin and the infiltration of eosinophils in nasal polyps. Methods:twenty specimens of nasal polyps and eight specimens of inferior turbinate mucosa as control were collected . Reverse transcription-PCR (RT -PCR )was used to investigate IL-5 and eotaxin mRNA expression. Immunohistochemical staining for eotaxin and IL-5 protein and HE staining for eosinophils were performed in nasal polyps and inferior turbinate mucosa. Results: (1)The postive infiltrating of eosinophil accounts for 75% in twenty cases of nasal polyps. (15 cases). It is higher than that 12.5% in normal matched control(1 case). And the infiltration ratio of eosinophils is 21.6±4.33 and is higher than that 1.15±0. 68 in the normal matched control(P〈0.01). (2) The protein expression of eotaxin and IL-5 in the nasal polyps are (Density of positive cell's) obviously more than that in the normal matchedcontrol (P〈0. 01). (3) Eotaxin and IL-5 mRNA expression in the nasal polyps are obviously more than that in the normal matched control(P〈0.05). (4) The expression of eotaxin and IL-5 in nasal polyps are correlated to the eosinophil infiltrating(P 〈 0 . 01) ,and there is a correlation between IL-5 and eotaxin (P〈0. 05). Conclusion: eotaxin and IL-5 content rise obviously in nasal polyps, and are correlated to the eosinophil infiltrating, suggesting that eotaxin and IL-5 are important chemotatic factor of eosinophil that take part in the pathological change of nasal polyps , and Strengthening some function of this cell in the focus and have participated in pathogenesis of the nasal polyp. And there is a correlation between them.
