重庆垫江牡丹皮HPLC指纹图谱研究(Ⅳ)——不同部位的比较及其鉴别意义

Study on the HPLC Fingerprint of Paeonia suffruticosa Cultivated in Chongqing Dianjiang County (Part Ⅳ): Comparison of Different Parts of Paeonia suffruticosa and Its Identification Significance

目的:比较重庆垫江药用牡丹根不同取材部位高效液相色谱(HPLC)指纹图谱的异同,为科学评价与有效控制牡丹皮质量提供新的依据和技术手段。方法:利用HPLC法(梯度洗脱)测定重庆垫江产牡丹根加工成的原丹皮、刮丹皮样品,并与全根和加工产生的木心、栓皮进行比较;采用国家药典委员会制定的《中药色谱指纹图谱相似度评价系统》(2004A版)专用软件进行自动匹配、相似度计算和色谱数据分析。结果:同株牡丹根各取材部位样品指纹图谱共有峰达15个,总峰面积、共有峰面积之和均为栓皮>全根>原丹皮>木心>刮丹皮。不同部位各色谱峰的相对峰面积差异较大,除原丹皮与刮丹皮外各样品间的相似度均<0.9。结论:牡丹根不同部位的指纹图谱存在显著差异,通过指纹图谱可以区别原丹皮和刮丹皮,并可以鉴别木心是否抽出,从而判断牡丹皮样品的纯度。加工过程对牡丹皮质量影响很大,采挖牡丹根后应及时抽去木心加工成牡丹皮,建议保留栓皮,加工成原丹皮。加工过程中产生的木心含有牡丹皮的主要有效成分丹皮酚、芍药苷及其它多种化学成分,应进行综合利用。

OBJECTIVE： To compare HPLC fingerprints of different parts of Paeonia suffruticosa cultivated in Dianjiang county so as to provide a new reference basis and technical method for the scientific appraisal and the effective control of the quality of P.suffruticosa.METHODS： Unshaved P.suffruticosa and shaved P.suffruticosa from Chongqing Dianjiang county were determined by HPLC （gradient elution） and were compared with that of root,duramen and cork.The software ＂Similarity Evaluation System for Chromatograph Fingerprint of TCM＂ （2004A version） issued by National Pharmacopoeia Committee of China was used for automatic matching,computation of similarities and chromatograph data.RESULTS： There are 15 common peaks in HPLC fingerprints of different parts of P.suffruticosa.The order of total peak area and common peaks were as follows： duramen,root,original P.suffruticosa,cork and shaved P.suffruticosa.There were striking differences among relative peak area of HPLC fingerprints of different parts of P.suffruticosa.The similarity of samples expect for unshaved P.suffruticosa and shaved P.suffruticosa were all lower than 0.9.CONCLUSION： There are significant differences among HPLC fingerprints of different parts of P.suffruticosa.HPLC fingerprints can be used for the identification of unshaved P.suffruticosa to shaved P.suffruticosa and the examination of purity of P.suffruticosa by testing the place of duramen.The quality of P.suffruticosa is influenced by producing process.Duramen is drawn out from original plant to produce P.suffruticosa.The cork of P.suffruticosa is maintained during process to prepare original P.suffruticosa.The duramen contains paeonol,peoniflorin and other chemical components,which also can be applied.
