摘要
目的:探讨复方栀黄颗粒治疗复发性口腔溃疡(ROU)的免疫作用机理。方法:用健康兔口腔黏膜匀浆作抗原制备兔ROU模型。将36只家兔随机分为空白组、模型组、左旋咪唑组、复方栀黄颗粒高剂量组(高剂量组)、复方栀黄颗粒中剂量组(中剂量组)、复方栀黄颗粒低剂量组(低剂量组)6组,每组6只。除正常对照组外,余组均造模。以健康家兔的口腔黏膜组织匀浆为抗原进行免疫注射制作家兔复发性口腔溃疡模型。造模结束后分别给药,疗程为15 d。给药期间定时观测动物口腔溃疡情况,末次给药后分别从家兔耳缘静脉采血(4 mL)并检测血中抗口腔黏膜抗体滴度,然后在麻醉下取血并检测T淋巴细胞亚群。结果:模型组T淋巴细胞亚群检测结果与空白组比较差异有统计学意义;左旋咪唑组与大剂量组、中剂量组、低剂量组均能提高CD4/CD8的比值,与模型组比较差异有统计学意义;左旋咪唑组能够提高CD4细胞数量的水平,与模型组比较差异有统计学意义;而高剂量组、中剂量组、低剂量组不仅能提高CD4细胞数量的水平,还可降低CD8细胞数量的水平,与模型组及左旋咪唑组比较差异有统计学意义;高剂量组、中剂量组、低剂量组与左旋咪唑组均能降低抗口腔黏膜抗体滴度;与模型组比较差异有统计学意义。结论:复方栀黄颗粒治疗ROU的免疫学机理可能是对细胞免疫、体液免疫的双向调节作用。
Objective:To explore mechanism of compound Zhihuang granule on rabbit with recurrent aphthous ulcer.Methods:36 male rabbits were divided into blank control group、model group、levamisole hydrochloride group、compound Zhihuang granule high dosage group、middle dosage group and low dosage group at random.Recurrent aphthous ulcer rab-bits were made by injecting with oral mucosa membrane of normal rabbits.15 d after treatment,the antibody titre resisting oral mucous membrane and T lymphocyte subsets in serum were examined.During the course of treatment,the change of oral ulcer was observed.Results:Compared with the model group,percentages of total T cells(CD3 T cells),CD4 T cells in peripheral blood were significantly decreased,percentage of CD8 was markedly increased;compared with model group,ratio of CD4 cells to CD8 cells from peripheral blood was significantly increased in levamisole hydrochloride group、high dosage group、middle dosage group and low dosage group;meanwhile percentage of CD4 was higher than that in model group;compared with model group and levamisole hydrochloride group,percentage of CD4 was markedly increased,percentage of CD8 was decreased in compound Zhihuang granule groups;compared with model group,the antibody titre resisting oral mucous membrane was decreased in levamisole hydrochloride group and compound Zhihuang granule groups.Conclusion: Compound ZhiHuang granule has a better therapeutic effect on recurrent aphthous ulcer.Its mechanism may be related to its two-way adjusting effects on both organic humoral immunity and cellar immunity.
