木醋杆菌纤维素对人成纤维细胞增殖活性的影响

摘要 目的:观察木醋杆菌纤维素对体外培养人成纤维细胞(Fbs)增殖活性的影响,探讨其促进创面愈合的可能机理。方法:采用消化分离法进行Fbs的原代培养,通过形态学和免疫组织化学法鉴定Fbs。将第6代纯化的Fbs随机分为与木醋杆菌纤维素敷料共同培养的敷料组织和空白对照组。培养24 h、48 h、72 h后,经MTT法检测每孔的光吸收值(OD值)。结果:木醋杆菌纤维素加入到Fbs中培养24 h、48 h、72 h后,经MTT法检测,木醋杆菌纤维素组与空白对照组OD值比较有显著性差异(P<0.05)。结论:木醋杆菌纤维素对Fbs的生长有明显促进作用,提示木醋杆菌纤维素敷料能明显增加Fbs数量,这可能与其可促进创面愈合的机理有关。 Objective:To investigate the effect of Acetobacter xylinum cellulose on the proliferation of fibroblast cells in vitro and to explore the mechanism of which Acteobacter xylinum promotes wound healing.Methods:Fibroblasts were obtained from the digested and aparted method of primary culture and identified by morphology and immunohistochemistry.The sixth purified fibroblasts were divided into two groups randomly and cultured with Acetobacter xylin cellulose dressing,and sham respectively.MTT methods was used to measure OD number of each hold after being cultured for 24 h,48 h,72 h respectively.Results:Fibroblasts cultured with Acetobacter xylin cellulose dressing,was compared with blank groups,OD numbers were obviously different(P0.05).Conclusion:Acetobacter xylinum cellulose can enhance the growth of fibroblast.Acetobacter xylinum cellulose dressing could increase the numbers of fibroblast obviously,and it may be beneficial to granulation tissue formation during wound healing.
