摘要
以甜瓜嫩芽叶尖为材料,采用去壁-火焰干燥法进行染色体制片,发现叶尖制片要求与根尖基本一致.利用所确定的制片程序,从随机选取的27个叶尖样品中检测出两个四倍体,一个嵌合体.除根尖和卷须之外,用叶尖为材料进行染色体制片,为染色体小的葫芦科作物的细胞学研究提供新的方法.
In this article,we studied the chromosome number of Cucumis melo using the leaf tips of immature buds via wall degradation and hypotonic treatment.We detected two tetraploids and a chimera from 27 randomly selected samples.As using the leaf tip for the preparation of chromosome materials was more convenient than using the root tip and the tendril,a new cytological method has been developed for cytological studies of cucurbitaceous crops which have small chromosomes and it is recommended for application in further ploidy breeding of C.melo.
