赖氨酰氧化酶样基因3在人皮肤成纤维细胞株中的表达

Expression of human lysyl oxidase-like gene 3 in human fibroblast

目的研究体外培养的人皮肤成纤维细胞株中赖氨酰氧化酶样基因3(lysyloxidase-likegene3,LOXL3)的表达模式。方法提取不同生长阶段的人皮肤成纤维细胞株的不同组分,用NorthernBlot和WesternBlot分别从mRNA和蛋白水平分析LOXL3表达,并以免疫荧光方法观察LOXL3的分布部位。结果 LOXL3mR-NA和蛋白含量在细胞汇合前低于细胞汇合期,汇合后最高,且LOXL3分布在整个细胞浆和细胞核。结论 LOXL3的表达量在细胞生长的不同阶段有所不同,可能与其功能有关。

Objective To study the expression pattern of Human Lysyl Oxidase-Like Gene 3（LOXL3）in cultured human skin fibroblast cells at different stages of confluncy.Methods The levels of LOXL3 mRNA and protein were detected by Northern and Western blot analyses,and the distribute pattern of LOXL3 in cells was observed through immunofluorescence technique.Results At different stages of cell culture,Northern and Western blot analyses showed a higher steady-state LOXL3 mRNA and protein level at 3 day after confluncy than confluncy and pre-confluncy.Immunofluorescence results showed that LOXL3 was localized in entire cytoplasm and nucleus.Conclusion These data demonstrate that the LOXL3 has different temporal distribution in human skin fibroblast cells.This might implicate the unique functions of LOXL3 in skin development.
