RUNX3在人类原发性肝细胞癌中的表达 被引量:3

Expression and clinical significance of Runt-related transcription factor 3 genes in primary hepatocellular carcinoma

摘要 目的探讨RUNX3基因在人类肝细胞性肝癌中的表达差异。方法采用免疫组化法和蛋白免疫印迹(WesternBlot)及逆转录-多聚合酶链反应(RT-PCR)技术,对临床肝细胞性肝癌50例石蜡标本和43例新鲜标本及其癌旁组织中RUNX3基因的表达进行检测。结果免疫组化结果表明RUNX3在肝细胞性肝癌组织中的表达率显著低于癌旁组织,两组比较差异有统计学意义(P<0.05);WesternBlot结果显示RUNX3蛋白在肝癌组织标本中的光密度值明显低于在癌旁组织中的光密度值,两组比较差异有统计学意义(P<0.05);RT-PCR结果显示RUNX3mRNA在肝癌组织表达率低于癌旁组织表达率,两者比较差异有统计学意义(P<0.05)。RUNX3蛋白及mRNA的表达与肿瘤分化程度有关(P<0.05),而与患者性别、年龄、HBsAg、AFP均无明显关系(P>0.05)。结论 RUNX3的表达下调与HCC的发生密切相关,可能作为抑癌基因在肝癌的发生发展中起作用。 Objective To investigate the expression and clinical significance of Runt-related transcription factor 3 gene(RUNX3)in human hepatocellular carcinoma.Methods Immunohistochemistry,Western Blot and Reverse transcription-PCR(RT-PCR)were used to detect RUNX3 mRNA and protein expression level of samples of primary hepatocellular carcinoma and corresponding para-cancer tissue.Results Immunohistochemistry showed that the expression rate of RUNX3 protein in hepatocellular carcinoma was significantly lower than that in corresponding para-cancer tissue(P0.05);Western Blot showed that the expression of RUNX3 protein in hepatocellular carcinoma was significantly lower than that in corresponding para-cancer tissue(P0.05);RT-PCR showed that the expression rate of RUNX3 mRNA in hepatocellular carcinoma was significantly lower than that in corresponding para-cancer tissue(P0.05),The depression of RUNX3 was correlated with pathologic differentiation(P0.05).Conclusion The down-regulated expression of RUNX3 gene in human hepatocellular carcinoma suggest that RUNX3 gene may play an important role in the development and progression of hepatocellular carcinoma as a suppressor of hepatocellular carcinoma.
