摘要
目的:观察五子衍宗丸含药血清对活性氧致大鼠精子鞭毛超微结构损伤的保护作用。方法:采用次黄嘌呤(0.6mM)-黄嘌呤氧化酶(0.05U/mL)活性氧发生体系,收集大鼠附睾精子,体外与活性氧及五子衍宗丸含药血清(低、中、高剂量)共孵育15min和60min,采用环境扫描电镜观察精子鞭毛超微结构的变化。结果:与活性氧组共孵育60min后,部分精子鞭毛质膜连续性中断,部分膜脱落,鞭毛暴露。各浓度五子衍宗丸含药血清(低、中、高剂量组)组精子鞭毛质膜完整。结论:五子衍宗丸含药血清体外对活性氧造成的精子鞭毛质膜损伤具有一定的保护作用。
Objective:To study the protective effect of Wuziyanzong(WZYZ)pill medicated serum on rat sperm flagellum ultrastructure damaged by reactive oxygen species (ROS).Methods:ROS was generated by hypoxanthine (0.6 mM)-xanthine oxidase (0.05 U/mL) system and then incubated with rat sperm flagellum and WZYZ medicated serum (low,medium and large dosage ) for 15 and 60 min. Surface ultrastructure of sperm was observed by environmental scanning electron microscopy (ESEM). Results:After incubation with ROS for 60 min,considerable changes were observed with the interruption,disruption and exfoliation of flagellum plasma membrane. While the flagellum plasma membrane of sperm in WZYZ medicated serum groups were intact. Conclusion:WZYZ pill medicated serum has a protective effect on the rat sperm flagellum ultrastructure damage induced by ROS.
