摘要
目的构建一种带有绿色荧光蛋白(GFP)和潮霉素磷酸转移酶和HSV-TK的融合基因(HyTK基因)的新型腺病毒载体,用于简便、快速地了解自杀基因在恶性肿瘤中的转染效率和提高疗效。方法利用脑炎心肌炎病毒的内部核糖体进入位点(internalribosomeentrysite,IRES),将GFP的cDNA与HyTK真核表达载体重组,构建获得含GFP和HyTK基因真核表达载体,在Lipofectin介导下转染COS-7细胞,检测其表达后进一步克隆获得含GFP和HyTK基因双顺反子腺病毒载体。结果构建含绿色荧光蛋白和HyTK基因双顺反子腺病毒载体,显示该载体可获得HyTK基因及GFP的良好表达。结论含绿色荧光蛋白和HyTK基因双顺反子腺病毒载体的构建为HyTK基因治疗恶性肿瘤用于临床提供了新的治疗工具及随访检测手段。
Objective\ In order to facilitate the suicide gene delivery into neoplasms, a chimeric gene of HSV TK and green flurescent protein(GFP) was constructed.\ Methods\ To construct this kind of adenoviral vector,method of molecular cloning was used.\ The internal ribosome entry site(IRES) which could coordinate expression of two genes in a single vector was optioned.\ Results\ A polycistronic adenoviral vector carring GFP and hygromycin phosphotransferase thymidine kinase fusion(HyTK) gene was successful constructed.\ Conclusion\ This new kind of adenoviral vector could serves as a new tool and method for neoplasms gene therapy.
