通用引物检测宫颈脱落细胞中病毒的临床应用

摘要 目的:探讨通用引物检测多种病毒的PCR方法。方法:应用聚合酶链反应-酶谱法(PCR-EC)和疱疹病毒通用引物,检测宫颈脱落细胞单纯疱疾病毒Ⅰ型(HSV—Ⅰ)、单纯疱疹病毒Ⅱ型(HSV—Ⅱ)、爱泼期坦—巴尔病毒(EBV)和人5细胞病毒(HCMV)。结果:健康人(20例)和宫颈糜烂患者(34例)。HCMV阳性检出率分别为10.0%和41.2%,两者差异显著(P<0.05);HSV—Ⅰ分别为5.O%和及14.7%;HSV—Ⅱ分别为1l%和20.6%;FBV分别为15.0%和17.6%;可检测到1fg的DNA。结论:通用引物PCR-EC检测HCMV、HSVⅠ、HSVⅡ、EBV,具有敏感、特异、便捷、快速、经济的特点,对病毒感染早期快速诊断及流行病学研究提供了科学的方法。 Objective: To study a PCR mothed which can detect more than one kind of virus infection.Methods: Polymerase chain reaction-endonuclease cleavage and single pair of ollgomucleotide primers selectedwithin a highly conserved region of DNA polymerase gene of the herpesvirus were used in detecting human cytomegalovirus (HCMV),herpes simplex virus Ⅰ,Ⅱ (HSV Ⅰ,HSV Ⅱ) and Eptein-Barr virus (EBV). Results: Twenty healthy women and 34 cervicitis were included in the study. The POsitive rates of HCMV inhealthy and cervicitis were 10. 0% and 41. 2% respectively (P<0' 05). The POSitive rates of HSV I,HSV E,EBV was 5. 0%, 10. 0%, 15. 0% in healthy and 14. 7 %, 20. 6%, 17. 6% in cervicitis,respectively. ~fusion:Detcting HCMV,HSV I,HSV E and EBV by single Pair of oligonucleotide primers selected within a highlyconserved region DNA polymetase gene Of the herpesvirus PCR-EC is sensitive,spesific,rapid and ecorlodric. Itprovided a scientific method in early diagnosis Of virus infection and epidentiology study.
