摘要
本文对花生、河虾中的主要过敏原蛋白进行分离纯化并制备抗体,同时探索胶体金作为免疫佐剂的可能性。结果表明,经电泳检验纯化后的过敏蛋白纯度超过90%,胶体金能够显著提高大白兔体内抗花生、河虾过敏蛋白的水平,花生与胶体金组的平均效价比弗氏佐剂组提高了一倍,其Ic50为34.28 ng/mL;河虾与胶体金组的平均效价比弗氏佐剂组提高了四分之一,其Ic50为43.95 ng/mL。该研究成功获得花生、河虾中主要过敏原组分及其抗体,且胶体金有可能开发为新一代免疫佐剂。
The major allergen protein from peanuts and freshwater shrimp were separated and purified as well as prepared their correspinging antibodies.The electrophoresis tests showed that the purity of allergens protein exceeded 90%.The quality of rabbits antiserum can be significantly improved while using colloidal gold as an immune adjuvant.The value of peanuts antibody with colloidal gold doubled compared with Freund's adjuvant,its Ic50 is 34.28 mg/L.Meanwhile,the value of freshwater shrimp and colloidal gold group were 1.25-fold compared with Freund's adjuvant and its Ic50 is 43.95 mg/L.The results showed that colloidal gold could become a new generation of adjuvant.
