摘要
目的研究高良姜水提物(AOWE)对SD大鼠尿液中内源性物质代谢的影响。方法给予SD大鼠ig0.5142g·kg-1AOWE,连续30d,收集尿液,采用LC-MS/MS分析尿液中内源性代谢物;通过标记物的变化,分析AOWE对尿液中内源性物质的影响。结果给药第8天,AOWE对大鼠尿液中代谢物的影响最为显著;OSC-PLS-DA分析有16种物质被视为可能生物标记物;与空白组比较,高良姜组大鼠的尿液中SM[d18:0/24:1(15Z)]、γ-linolenyl carnitine等物质浓度升高,PA[18:0/18:2(9Z,12Z)]、CerP(d18:1/22:0)等的浓度降低。结论高良姜的水提物可显著影响细胞信号传导类物质的代谢。
OBJECTIVE To investigate the changes of endogenous metabolites concentrations in the urine of rats administrated with Alpinia officinarum water extracts.METHODS The animals were administrated with Alpinia officinarum water extracts for 30 days.Urine was collected at 0,1,8,15,22 d and 29 d after administration respectively and analyzed by LC-MS/MS.The acquired data were analysed by OSC-PLS-DA.RESULTS After administration for 8 days,endogenous metabolites concentrations in the urine of rats changed apparently.Through OSC-PLS-DA analysis,16 biomarkers were found.The identified biochemical effects of these biomarkers were associated with the pharmacological actions of Alpinia officinarum which included increasing concentrations of SM(d18:0 /24:1(15Z)),Gamma-linolenyl carnitine and decreasing concentrations of PA(18:0 /18:2(9Z,12Z)),CerP(d18:1 /22:0).CONCLUSION The administration of Alpinia officinarum water extracts could have changed the cell signals of rats significantly.
