摘要
目的研究龙葵碱对人宫颈癌细胞系HeLa的凋亡作用及其抑癌机制。方法采用噻唑蓝(MTT)法检测龙葵碱对He-La细胞生长的影响;通过Hoechst 33258染色及DNALadder研究龙葵碱诱导HeLa细胞凋亡的作用;RT-PCR法检测龙葵碱对环氧合酶(COX-2)表达的影响。结果龙葵碱可剂量依赖性地抑制HeLa细胞生长,作用48h时,IC50=260μg·mL-1;可诱导HeLa细胞凋亡,荧光染色可见凋亡小体,DNALadder显示典型的细胞凋亡梯形条带;可降低COX-2的表达水平。结论龙葵碱可抑制人宫颈癌细胞HeLa的增殖,诱导细胞凋亡,这可能是龙葵碱抑癌作用的机制之一。
OBJECTIVE To investigate the apoptosis-inductive effects of solanine on human cervical cancer cell line HeLa in vitro.METHODS HeLa cells were treated with solanine of different concentrations for 48 h and the cell viability was assessed by MTT assay.The morphological changes of HeLa cells undergoing apoptosis were visualized by fluorescent staining with Hoechst 33258.Semi-quantitative RT-PCR was used to assess the expression of COX-2.RESULTS Solaine inhibited the HeLa cells'growth in a dosage dependent manner with IC50 value of 260 μg·mL-1 when administrated for 48 h.Fluorescence staining with Hoechst 33258 showed nuclear condensation and apoptosis bodies in HeLa cell.DNA fragmentation assay demonstrated typical ladder pattern of isolated genomic DNA on agarose gel electrophoresis.Expression level of COX-2 decreased when treated with solanine.CONCLUSIONS Solanine could inhibit cell growth and induce cell apoptosis in HeLa cells,which may be one of the mechanisms of solanine's antitumor effect.
