摘要
本文报道了应用混合纤维素酯微孔滤膜作载体,进行斑点酶联免疫吸附法测定(Dot-Enzyme Linked lmmuno Sorbert Assay,D-ELISA)赭曲霉毒素A (Ochratoxin A,OA的试验研究。试验中应用作者偶合的OA-BSA抗原,制备的兔抗OA抗体,经稀释200×、400×、800×、1600×后。对不同浓度的纯OA进行试验。用D-ELISA间接法测得的抗OA抗体效价为1∶800,OA最小检出量为0.0025μg/ml,具有敏感性高,特异性强,简便易行,结果易于判断并能永久保留等优点。
Determination of ochratoxin A (OA) with Dot-enzyme linked immunosorbent assay(D-ELISA) was conducted using microporus filter membrane made of the mixture of cellu-lose acetate and cellulose nitrate as carriers. The antibody to OA-BSA antigen made by theauthor was prepared from rabbit and then dilated 200×, 400×, 800×, 1600× to measurethe pure OA (made by the author) at different concentrations with indirect D-ELISA. Thetitre of the antibody to OA was 1:800 and the least amount of OA that could be measuredwas 0.0025μg/ml. The indirect D-ELISA was proved to be highly sensitive, stronglyspccialized, convenient and practicable, easy for judgement, and permanent for reservation.
