东北天南星体细胞胚胎发生及植株再生体系的优化 被引量:3

摘要 为建立东北天南星体细胞胚胎发生体系,以东北天南星幼叶为外植体,应用均匀设计法筛选其最适合的胚性愈伤组织及胚性细胞复合体诱导、体细胞胚胎发育及植株再生的培养基。结果表明,东北天南星叶片胚性愈伤组织及胚性细胞复合体诱导最适宜的培养基为MS+6-BA 2.5 mg/L+2,4-D 2.0 mg/L+IAA 0.1 mg/L,诱导率为98%;体细胞胚胎发育及植株再生最适宜的培养基为MS+6-BA 0.9 mg/L+NAA 0.1 mg/L+IAA 0.1 mg/L,体细胞胚萌发率为99.5%,萌发的胚20 d后全部发育成完整植株。 Young leaves of Arisaema amurense were used as explants to screen out the optimum media for embryonic callus and embryonic cell complex induction,somatic embryo development and plantlet regeneration by using the uniform design to establish a somatic embryogenesis system of Arisaema amurense.The results showed that the optimum medium for 98% induction rate of embryonic callus and embryonic cell complex was MS+6-BA 2.5mg/L+ 2,4-D 2.0mg/L +IAA 0.1mg/L,and the optimum medium for 99.5% emergence rate of somatic embryos was MS+6-BA 0.9mg/L +NAA 0.1mg/L +IAA 0.1mg/L and the intact plantlets could be successfully developed from the emerged embryos cultured on the same medium after 20d.The development process of somatic embryos was illustrated by morphological structure and ultra-structure of cultured materials with different development stages.
