丙戊酸钠对CIK细胞杀伤K562细胞活性的影响

摘要 目的观察丙戊酸钠(VPA)对细胞因子诱导杀伤(CIK)细胞杀伤K562细胞活性的影响。方法通过半定量RT-PCR、流式细胞术等方法检测经VPA诱导的K562细胞MICA的mRNA和蛋白表达情况。MTT法检测CIK细胞对经VPA诱导的K562细胞的杀伤活性。结果经VPA诱导的K562细胞的MICA-mRNA、细胞膜MICA表达增加;CIK细胞对经VPA诱导的K562细胞的杀伤活性增加,杀伤活性与K562细胞膜MICA表达呈正相关。结论VPA可以上调K562细胞MICA的表达,进而增加其被CIK细胞杀伤的敏感性。 Objective To investigate the effect of sodium valproate(VPA) on the sensitivity of K562 cells to the cytotoxicity of cytokine-induced killer(CIK) cells.Methods The mRNA and protein levels of MICA of K562 cells were measured by semi-quantitative RT-PCR and flow cytometry,respectively.The cytotoxicity of CIK cells against VPA-induced K562 cells was assessed using MTT assay.Results VPA treatment resulted in a significant increase in the MICA mRNA and membrane MICA expression as well as the sensitivity of K562 cells to the cytotoxicity of CIK cells.Moreover,the sensitivity of K562 cells to CIK cells was positively correlated with the membrane MICA expression.Conclusion VPA can increase the sensitivity of K562 cells to CIK cells,at least partially through the up-regulation of MICA expression.
