摘要
目的研究亚低温对易卒中型肾血管性高血压大鼠(RHRSP)复制的大脑中动脉闭塞(MCAO)模型的梗死灶大小及对BCL-2蛋白表达的影响。方法用环型银夹以双肾双夹法复制RHRSP,改良Longa’s线栓法制成MCAO缺血模型,冰袋法诱导亚低温,TTC染色测定梗死灶体积,免疫组化法、图象识别系统计算BCL-2蛋白表达强度。结果1亚低温能明显缩小梗死灶体积;2亚低温组BCL-2蛋白表达明显强于正常体温组。结论亚低温增强缺血后基因BCL-2的表达可能是它具有缺血脑保护作用机制之一。
s Objective To investigate the effects of mild hypothermia (MH) on expression of BCL-2 protein in a middle cerebral artery occlusion (MCAO) modle of stroke-prone renovascular hypertensive rate (RHRSP).Methods RHRSP were made by constricting bilaterally the renal arteries with silver clips.The MCAO was made by Longa's method. MH was induced systemically by ice bag to reduce the core body temperature of the animals and the rectal temperature was maintained at 32-34℃ for 3 hours.The infarct volume was measured by TTC staining.The expression of BCL-2 protein was assessed by immunostaining method.All datas were analyzed by SPSS for window package with significance level at 0.05. Results A significant increase in expression of BCL-2 protein in MH group was observed compared with that of control group.In the meantime,the infarct volume in MH group was smaller than that of control group. Conclusions The results show that MH will increase the expression of BCL-2 protein which may play a role in protection of ischemic neurons.
