Hep-2细胞中bFGF及其双受体系统mRNA的表达

摘要 目的:研究碱性成纤维细胞生长因子(bFGF)及其双受体系统串珠素(perlecan)和高亲和力酪氨酸激酶受体(FGFR)在人喉癌细胞Hep-2中的表达。方法:培养Hep-2和正常永生化表皮细胞系HaCaT细胞,应用RT-PCR方法检测bFGF、perlecan及FGFR1~4 mRNA的表达。结果:bFGF、perlecan、FGFR1、FGFR2及FGFR4 mRNA在Hep-2细胞中的相对表达量高于HaCaT细胞(t分别为17.540、19.684、13.371、12.692和12.586,P均<0.05)。2种细胞中均未检测到FGFR3 mRNA。结论:bFGF、perlecan及FGFR与喉癌细胞的生长关系密切。 Aim:To study the expressions of basic fibroblast growth factor (bFGF),perlecan and the four high-affinity FGF receptors(FGFR) in human laryngeal carcinoma cells.Methods:The expressions of bFGF,perlecan and FGFR1~4 mRNA in human laryngeal carcinoma cells Hep-2 were detected using techniques of semi-quantity RT-PCR.The normal cells,HaCaT cells,were the control.Results:FGFR3 mRNA was not detected in the two kinds of cells.The expression levels of bFGF,perlecan,FGFR1,FGFR2 and FGFR4 mRNA in Hep-2 cells were significantly higher than those in HaCaT cells (t was 17.540,19.684,13.371,12.692 and 12.586,P0.05).Conclusion:bFGF,perlecan and its high-affinity FGFR may play key roles in the growth of human laryngeal carcinoma cells.
