人脂肪间充质干细胞体外分化为视网膜细胞观察 被引量:4

摘要 目的:研究成人脂肪间充质干细胞(hAD-MSC)体外诱导分化为视网膜色素上皮细胞(RPE)、光感受器细胞和血管内皮细胞的情况。方法:分离、培养和鉴定hAD-MSC,用血管内皮生长因子和成纤维细胞生长因子诱导hAD-MSC向血管内皮细胞分化,并检测血管内皮细胞标志物血管内皮细胞Ⅷ因子(vWF)的表达情况。制备hAD-MSC体外诱导液,用诱导剂和诱导液共同诱导hAD-MSC向视网膜细胞分化,采用免疫荧光法检测hAD-MSC经诱导后表达光感受器细胞标志物视紫红质(Rhodopsin)和RPE细胞标志细胞角蛋白(Pan-CK)的表达情况。结果:免疫荧光法证实,分离培养的hAD-MSC经体外诱导后可表达RPE、光感受器细胞和血管内皮细胞的标志Pan-CK、Rhodopsin和vWF。结论:hAD-MSC经过体外诱导可以向视网膜RPE、光感受器细胞和血管内皮细胞分化。 Aim:To investigate the differentiation of human adipose derived-mesenchymal stem cell (hAD-MSC) into cells of retinal in vitro.Methods:hAD-MSC was isolated,cultured and identified. hAD-MSC was induced by VEGF and bFGF. Cell morphology and expression of vascular endothelial cell-specific marker vWF were studied. Rat retinal cells were isolated and used to produce the induced liquid.Immunofluorescence was used to identify the ability of hAD-MSC to be induced by taurine,retinoic acid and the induced liquid into cells expressing RPE-specific marker Pan-CK and photoreceptor-specific marker Rhodopsin in vitro.Results:hAD-MSC could be induced to express vWF,Pan-CK and Rhodopsin in vitro.Conclusion:hAD-MSC has the ability to be induced into vascular endothelial cells,RPE and photoreceptor cell in vitro.
