苯丁酸钠体外对YTMLC-90细胞生长的影响 被引量:1

摘要 目的:观察苯丁酸钠(PB)体外对人个旧肺鳞状细胞癌YTMLC-90细胞生长的影响。方法:YTMLC-90细胞分为6组,对照组不作任何处理,余5组分别给予250、500、1000、2000和4000mg/LPB,分别作用48、72和96h。采用MTT法观察各组细胞生长情况,计算细胞生长抑制率。另取YTMLC-90细胞分为3组:对照组、500和1000mg/LPB组,分别作用48和72h,采用流式细胞术检测3组细胞的细胞周期及凋亡情况。结果:不同质量浓度的PB作用48、72和96h,各组细胞的生长抑制率相比,差异均有统计学意义(F=672.413、1633.321和1196.548,P均<0.05)。PB作用48和72h后,各组细胞G0/G1期、S期与G2/M期细胞百分率及凋亡率比较,差异均有统计学意义(F48h=508.347、224.522、19.890和111.849,F72h=311.979、102.923、247.432和177.018,P均<0.05)。与对照组相比,PB作用后YTMLC-90细胞生长缓慢,细胞阻滞于G0/G1期,凋亡增加(P<0.05)。结论:PB可抑制YTMLC-90细胞的生长,使其阻滞于G0/G1期,凋亡增加。 Aim:To study the effects of sodium phenylbutyrate(PB) on human lung squamous cancer cell line YTMLC-90.Methods:YTMLC-90 cells were allocated into 6 groups. The control group was not treated,and the other 5 groups were given 250,500,1 000,2 000,and 4 000 mg/L PB,and treated for 48,72,and 96 h.MTT assay was used to evaluate the inhibitory effects of PB on the cells.YTMLC-90 cells were allocated into 3 groups.The control group was not treated,and the other 2 groups were given 500 and 1 000 mg/L PB for 48 and 72 h,respectively. The flow cytometry was applied to detect the apoptotic rate and alteration of cell cycle.Results:After being treated for 48,72,and 96 h,the cell growth inhibition rate among the 6 groups had significant differences (F=672.413,1 633.321,and 1 196.548,all P0.05).After being treated for 48 h,the fraction of G0/G1 phase cells,S phase cells,and G2/M phase cells,and the apoptosis rate among the 3 groups had significant differences (F48 h=508.347,224.522,19.890,and 111.849;F72 h=311.979,102.923,247.432,and 177.018,all P0.05). PB could inhibit the growth of YTMLC-90 cells and most of the cells were arrested at G0/G1 phase(P0.05).Conclusion:PB could inhibit the growth of YTMLC-90 cells in vitro. The inhibitory effects of PB on YTMLC may through the increasing of G0/G1 fraction and apoptosis induction.
